Advances in Alzheimer's disease (AD) research have been challenging and without major breakthroughs in understanding its pathological basis. The reigning hypothesis suggests AD is the result of extracellular amyloid deposition that seed to form amyloid plaques, which then grow and kill neighboring neurons. However, there are several inconsistencies with this hypothesis, not to mention the inability to show clinical benefit in several failed clinical trials by pharmaceuticals (i.e., from Pfizer, Eli Lilly, etc.), and it is in the field's best interest to explore and test multiple hypotheses for pathology rather than drive the majority of research on this single amyloid theory. Reviewing many scientifically peer-reviewed publications, this book describes the "Inside-Out" hypothesis on how amyloid escapes the circulatory system through a dysfunctional blood-brain barrier to bind to the alpha 7 nicotinic acetylcholine receptor on pyramidal neurons. Over time, excessive amounts of amyloid appear to be internalized, resulting in neuron death and lysis. This simple mechanism readily explains plaque composition, size, shape, and location. Based on the current direction of research in the field, this hypothesis appears years from any research and development.

Описание: With the discovery of an ever increasing number of biogenetic polypeptides in the autonomic nervous system, it is evident that nervous transmission is controlled and modified not only by a series of classical transmitters but also by a variety of co-transmitters and modulators. Purifi- cation, characterization and synthesis of neuropeptides and peptide families allows the application of specific anti- bodies for radioimmunological, immunohistochemical and immunocytochemical localization. Transmitter interactions and phenotypic neuronal plasticity can be analyzed by pharmacological or surgical manipulations. Biochemical and autoradiographic determination of specific membrane receptors enables the recognition of peptide target struc- tures. In addition, neurophysiological techniques in combi- nation with histochemical methods result in functional interpretations. With regard to somatic manifestations of autonomic nervous disturbances these studies will offer fundamental insights for clinical medicine. This volume contains the refereed proceedings of recent investigations on histochemistry and cell biology of auton- omic neurons and paraganglia which were presented at an international colloquium on nervous transmission in Heidelberg in September 1986. The colloquium was origi- nally planned to be held under the presidency of Olavi Eranko but was continued by the editor after his death. It was sponsored by the German Research Foundation and took place on the occasion of the 600th anniversary of the Heidelberg University "Ruperto Carola" as part of the pro- gram in the international "Wissenschaftsforum". Hopefully, the information provided in this book will stimu- late new ideas and research strategies in the field of neu roscience.

Описание: The visual system of insects has attracted histologists for a long time. We have detailed histological studies of the visual systems of Diptera, Hymenoptera and Odonata dating from the last century: Leydig's (1864) study on optical ganglia of insects, Ciaccio's (1876) on the fine structure of the first ganglion in the mosquitos and Hickson's (1885) giving for the first time an exact description of the three optical ganglia of Musca. From 1896 several papers appeared using neuro- histological methods, mainly Golgi techniques and methylene blue staining. In 1896 Kenyon published his work on the bee-brain and in 1897 described the relationships of neurons in the optic ganglia with a modified Golgi method. Another work, by Jonescu (1909), should be mentioned: "Vergleichende Unter- suchungen am Gehirn der Honigbiene." In the same year Cajal's findings on the optic ganglia of the fly were published; then in 1915 Cajal and Sanchez wrote a definitive monograph on the neural elements and their connections in the same animal that remained the main reference in this field for decades. In both works Golgi techniques were used. Since then there have been only a few new publications on the subject: (Gribakin, 1967; Perrelet and Baumann, 1969 a, b; Perrelet, 1970; Varela and Porter, 1969; Skrzipek and Skrzipek, 1971, 1973; Grundler, 1972: Snyder, Menzel and Laughlin, 1973). They deal mainly with the receptors of the retina.
