Описание: The hepatocarcinogenicity of aflatoxin BI in rats, ducks and rainbow trout has been convincingly demonstrated (1). In contrast, adult mice appear to be resistant to the toxic and carcinogenic effects. Newborn and infant mice are more sensitive to the effects of different hepatocarcinogens (2,3) and also to AFB, which induced hepatomas in newborn C57BL/6 x C3H FI I mice (4). A singl dose model of aflatoxin hepatocarcinogenesis was tested in infant C57BL/6 mice. AFBI was administered i.p. on day 7 after birth and the following data were investigated: 1) Changes in the activities of microsomal cytochrome P-450 dependant enzymes and conjugating enzymes in the course of the 20-month-Iong aflatoxin hepa- tocarcinogenesis study. At intervals of 6,8,12,18 and 20 months a ter AFBI administration, the concentrations of cyt P-450 andb and the acti- 5 vities of NADPH-, NADH-cyt c reductase, AOH, ADM, EH and GST were deter- mined. 2) Effect of continually administered diethyldithiocarbamate on the course of aflatoxin carcinogenesis. MATERIALS AND METHODS Aflatoxin BI was prepared biosynthetically in our laboratory and was checked for purity chromatographically, spectrophotometrically and by NMR.

Описание: N-nitroso-compounds form an important class of carcinogenic chemicals which may be of environmental importance (Druckrey et al., 1967; Magee et al., 1976; IARC, 1978). The mechanism by which these carcinogens initiate neoplastic growth is not well understood but there is substantial evidence favoring the hypothesis that the simple dialkylnitrosamines and alkylnitrosamides act by means of their conversion to al- kylating agents. Alkylating species are generated from the nitrosamines by metabolic activation and from the nitrosamides by chemical decomposition at physiological pH. It is widely believed that DNA is the critical alkylation target. Alkylation takes place at at least 12 sites within the DNA molecule (Lawley, 1976; Singer, 1976; Pegg, 1977a). Al- though alkylation at the 7-position of guanine is the most extensive reaction with the DNA bases and provides a reliable measurement of the degree of interaction with the carcinogen (Swann and Magee, 1968, 1971), recent experiments have suggested that alkylation of oxygen atoms may be the critical reaction (Goth and Rajewsky, 1974; Margison and Kleihues, 1975; Nicoll et aI., 1975; Pegg, 1977a). These laboratories have observed a correlation between the production and persistence of 06-alkyl- guanine and the occurrence oftumors in a variety of organs of rodents exposed to N-ni- troso-carcinogens. It has, therefore, been suggested that the ability of tissues to catalyze the removal of 06-alkylguanine from DNA may provide a protective mechanism against carcinogenesis.

Описание: Stochastic processes often pose the difficulty that, as soon as a model devi- ates from the simplest kinds of assumptions, the differential equations obtained for the density and the generating functions become mathematically formidable. Worse still, one is very often led to equations which have no known solution and don't yield to standard analytical methods for differential equations. In the model considered here, one for tumor growth with an immunological re- sponse from the normal tissue, a nonlinear term in the transition probability for the death of a tumor cell leads to the above-mentioned complications. Despite the mathematical disadvantages of this nonlinearity, we are able to consider a more sophisticated model biologically. Ultimately, in order to achieve a more realistic representation of a complicated phenomenon, it is necessary to examine mechanisms which allow the model to deviate from the more mathematically tractable linear format. Thus far, stochastic models for tumor growth have almost exclusively considered linear transition probabilities.
