Описание: The genetic information contained in a cell needs the appropriate environment to express itself, not only the intracellular environment but also the extracel- lular one. The latter is provided to a great extent by the molecules which constitute the extracellular matrix. On the one hand, the matrix creates inter alia the right pH and osmotic envi- ronment and allows the diffusion of messengers targeting the cell membrane; on the other hand, it has a mechanical effect whose relevance began to be understood 28 years ago. Basically, the messages that reach the cell and are then transported to the genome depend on molecular conformational flexibility. Molecular structures usually prevail because they represent states of minimum potential energy cre- ating energy barriers which are activated through conformational changes. From the periphery to the nucleus the information flows through the activa- tion of energy barriers. The tools used to switch from low-energy to high- energy molecular configurations are: the binding of ligands to their receptors, gradients of electrochemical potential created by ion pumps, Ca2] mobiliza- tion, and phosphorylation and dephosphorylation. Variation in molecular con- figuration through molecular binding is in itself sufficient to trigger ion pumps and activate kinases and phosphatases. This is one aspect of the mechanical role of the extracellular matrix dealt with herein: the induction of molecular and supramolecular conformational modifications through interactions with the cell membrane, which promote the transduction and centripetal progression of signals.

Описание: This book encompasses the exciting developments and challenges in the fast-moving and rapidly expanding research field of single-molecule kinetic analysis of cell signaling that promises to be one of the most significant and exciting areas of biological research for the foreseeable future. Cell signaling is carried out by complicated reaction networks of macromolecules, and single-molecule analyses has already demonstrated its power to unravel complex reaction dynamics in purified systems. To date, most of the published research in the field of single-molecule processes in cells, focus on the dynamic properties (translational movements of the centre of mass) of biological molecules. However, we hope that this book presents as many kinetic analyses of cell signaling as possible. Although single-molecule kinetic analysis of cellular systems is a relatively young field when compared with the analysis of single-molecule movements in cells, this type of analysis is highly important because it directly relates to the molecular functions that control cellular behavior and in the future, single-molecule kinetic analysis will be largely directed towards cellular systems. Thus, we hope that this book will be of interest to all those working in the fields of molecular and cell biology, as well as biophysics and biochemistry.

Описание: Polyamines are organic cations found in all eukaryotic cells and intimately involved in, and required for, distinct biological functions. An increasing body of evidence indicates that the regulation of cellular polyamines is a central convergence point for the multiple signaling pathways driving various cellular functions. Over the last decade, considerable progress has been made in und- standing the molecular functions of cellular polyamines. These significant findings provide a fundamental basis to not only define the exact role of polyamines in physiology, but also to develop new therapeutic approaches for cancers and other diseases. The major objective of this book is to provide a timely and long lasting guide for investigators in the fields of polyamines, physiology, pharmacology, and cancer research. It will provide a foundation based on research and address the potential for subsequent applications in clinical practice. Polyamine Cell Signaling: Physiology, Pharmacology, and Cancer Research is divided into four main parts: Part I: Polyamines in Signal Transduction of Cell Proliferation Part II: Polyamines in Cellular Signaling of Apoptosis, Carcinogenesis, and Cancer Therapy Part III: Polyamines in Cell Motility and Cell-Cell Interactions Part IV: Polyamine Homeostasis and Transport This book not only covers the current state-of-the-art findings relevant to cellular and molecular functions of polyamines, but also provides the underlying conceptual basis and knowledge regarding potential therapeutic targeting of polyamines and polyamine metabolism. These points are addressed by int- nationally recognized experts in their contributions to this book.

Описание:

Mitochondria have traditionally been associated with metabolic functions; however recent research has uncovered a central role for these organelles in cell signaling, cell survival, and cell death. Mitochondrial dysfunction is a factor in a myriad of pathophysiological conditions, including age-related neurodegenerative disorders, cancer, metabolic syndrome, and cardiovascular disease. Mitochondrial Signaling in Health and Disease examines themes essential for the maintenance of the mitochondrial redox (reduction-oxidation) energy axis.

With contributions from an impressive cadre of internationally recognized scientists, the book discusses coordinated mitochondrial functions that regulate cell function by discrete signaling pathways. Topics discussed include:

  • Electron transport and energy production
  • Mitochondrial biogenesis and dynamics
  • Mitochondrial signaling
  • Apoptosis and autophagy
  • Pharmacology signaling
  • Epigenetic signaling: mitochondrial methylation and acetylation reactions

An essential resource for life and health scientists as well as pharmaceutical industry professionals, this volume highlights the importance of mitochondrial signaling and its role in establishing a harmonized communication between several cellular compartments. The information in this volume is critical to those developing mitochondrion-targeted therapies aimed at assuaging mitochondrial dysfunction or the specific factors contributing to its dysfunction.
