Описание: The 1st Euchem Symposium on the Chemical Synthesis of Antibiotics was held in Aussois in Savoie, France on May 2-6, 1988 thanks to the efforts of Prof. G. Lukacs, chairperson, and Prof. F. Arcamone and Prof. P. Welzel, vice-chairpersons of the International Organizing Committee. Following the great success of the Ist Symposium, the 2nd Symposium, which was chaired by Prof. Ohno, was held in Oiso, Japan in 1990. The 3rd Symposium, chaired by Prof. K. Krohn, was held in Kloster Banz, Germany in 1992. It has been decided that the 4th Symposium will take place in the United States in 1994 and be chaired by Dr. H. A. Kirst of Lilly Research Laboratories. The Euchem Symposium has come to serve as an important symposium where one can understand the development and the future directions of the chemical synthesis of antibiotics and other bioactive metabolites from microorganisms. Prof. G. Lukacs, in cooperation with Prof. M. Ohno, edited the book "Recent Progress in the Chemical Synthesis of Antibiotics" three years ago. Most of its contributors were the participants of the Aussois Symposium. This book attracted the attention of researchers specializing in organic synthesis and stimulated considerable progress in this field.

Описание: Antibiotics are among the most widely prescribed drugs in both human and veterinary medicine. When we started to teach our course on the science of antibiotics at the University of Pavia and the University of Milan, we realized that there was no book that presented the basic facts and concepts on all aspects of this diverse science.

Описание: Antibiotics are truly miracle drugs. As a class, they are one of the only ones that actually cure disease as opposed to most drugs that only help relieve symptoms or control disease. Since bacteria that cause serious disease in humans are becoming more and more resistant to the antibiotics we have today, and because they will ultimately become resistant to any antibiotic that we use for treatment or for anything else, we need a steady supply of new antibiotics active against any resistant bacteria that arise. However, the antibiotics marketplace is no longer attractive for large pharmaceutical companies, the costs of development are skyrocketing because of ever more stringent requirements by the regulatory agencies, and finding new antibiotics active against resistant strains is getting harder and harder. These forces are all combining to deny us these miracle drugs when we need them the most. I provide a number of possible paths to shelter from this perfect storm.

Описание: The International Society of Chemotherapy meets every two years to review progress in chemotherapy of infections and of malignant disease. Each meeting gets larger to encompass the extension of chemotherapy into new areas. In some instances, expansion has been rapid, for example in cephalosporins, pen icillins and combination chemotherapy of cancer - in others slow, as in the field of parasitology. New problems of resistance and untoward effects arise; reduction of host toxicity without loss of antitumour activity by new substances occupies wide attention. The improved results with cancer chemotherapy, es pecially in leukaemias, are leading to a greater prevalence of severe infection in patients so treated, pharmacokinetics of drugs in normal and diseased subjects is receiving increasing attention along with related problems of bioavailability and interactions between drugs. Meanwhile the attack on some of the major bacterial infections, such as gonorrhoea and tubercu losis, which were among the first infections to feel the impact of chemotherapy, still continue to be major world problems and are now under attack with new agents and new methods. From this wide field and the 1,000 papers read at the Congress we have produced Proceedings which reflect the variety and vigour of research in this important field of medicine. It was not possible to include all of the papers presented at the Congress but we have attempted to include most aspects of cur rent progress in chemotherapy.

Описание: Antibiotics: Challenges, Mechanisms, Opportunities details the contemporary challenges for discovery, optimization, and advancement of new antibiotics. It focuses on discovery not preclinical or clinical development. Given that the few classes of clinically significant antibiotics are all small molecules, this book focuses on the molecular structures and characteristics of both natural and synthetic antibacterial agents. Antibiotics is a valuable reference for microbiologists, bacterial physiologists, geneticists, pharmacologists, and drug developers. A chemocentric view of the molecular structures of antibiotics, their origins, actions, and major categories of resistance Antibiotics: Challenges, Mechanisms, Opportunities focuses on antibiotics as small organic molecules, from both natural and synthetic sources. Understanding the chemical scaffold and functional group structures of the major classes of clinically useful antibiotics is critical to understanding how antibiotics interact selectively with bacterial targets. This textbook details how classes of antibiotics interact with five known robust bacterial targets: cell wall assembly and maintenance, membrane integrity, protein synthesis, DNA and RNA information transfer, and the folate pathway to deoxythymidylate. It also addresses the universe of bacterial resistance, from the concept of the resistome to the three major mechanisms of resistance: antibiotic destruction, antibiotic active efflux, and alteration of antibiotic targets. Antibiotics also covers the biosynthetic machinery for the major classes of natural product antibiotics. Authors Christopher Walsh and Timothy Wencewicz provide compelling answers to these questions: • What are antibiotics? • Where do antibiotics come from?• How do antibiotics work? • Why do antibiotics stop working? • How should our limited inventory of effective antibiotics be addressed? Antibiotics is a textbook for graduate courses in chemical biology, pharmacology, medicinal chemistry, and microbiology and biochemistry courses. It is also a valuable reference for microbiologists, biological and natural product chemists, pharmacologists, and research and development scientists.
