Описание: This book on “Renin-Angiotensin System in Cardiovascular Disease” includes 25 chapters, which are organized in three sections, namely (i) modulatory aspects, (ii) pathophysiological aspects, and (iii) pharmacotherapeutic aspects. It includes an updated as well as comprehensive knowledge about molecular and cellular aspects for the role of the renin-angiotensin system (RAS) in the pathophysiology and therapy of cardiovascular diseases such as hypertension, atherosclerosis, ischemic heart disease, and heart failure. This book emphasizes the molecular and cellular mechanisms, signaling transduction pathways involved in the development of different cardiovascular diseases due to the prolonged activation of RAS. Furthermore, biochemical mechanisms are outlined for the inhibition of this system by the blockade of angiotensin converting enzyme as well as angiotensin II type 1 receptors in patients suffering from cardiovascular abnormalities. Since cardiovascular disease is the number one cause of death worldwide, leading to approximately 17.9 million deaths each year, there is a keen interest in understanding the pathogenesis and improving its therapy. In this regard, we can attest that this book provides ample information about essential components of RAS and their role in the development of cardiovascular disease. From the selection of recognized global experts in their area of investigation, this book can be seen to cover diverse cardiovascular aspects and molecular and cellular mechanisms of angiotensin II action for the development of different cardiovascular abnormalities. Описание: The first publications on the presence of renin, angiotensino- gen and angiotensin-like material in brain tissue appeared about ten years ago. This coincided with the discovery of new biological actions of angiotensin in the brain and with the reinvestigation of already known central angiotensin effects. The possibility that angiotensin could be generated within the brain itself had therefore exciting implications. With the recent popularity of brain neuropeptides, interest in this area is even increasing. The discovery of renin-like activity as well as angiotensino- gen and angiotensin in the brain has led, admittedly by inductive inference, to the hypothesis of the existence of a complete endogenous renin-angiotensin system in the brain.1 For several years there was ardent discussion whether such a renin angiotensin system existed in the brain or not. Inductive inference always contains "an irrational element or a creative intuition".2In view of the biological relevance of a brain angiotensin system, deductive testing of the hypothesis was a worthwhile scientific project taken up by a number of investigators As a matter of principle it is not given to science, to reach either truth or falsity, scientific statements can only attain contmous degrees of probability whose unat- tamable upper and lower limits are truth or falsity. However, several of the early singular statements and results turned out to be reproducible and acceptable. The theory has thus so far passed its test and we have found no reason to discard it.

Описание: Angiotensin-converting enzyme (ACE) is a well-known zinc-metallopeptidase that converts angiotensin I to the potent vasoconstrictor angiotensin II and degrades bradykinin, a powerful vasodilator, both for the regulation of vascular tone and cardiac functions. Other natural substrates of ACE were identified broadening the functions of this enzyme within different physiological contexts such as neuronal metabolism, hematopoiesis, digestion, and reproduction. In this context, ACE has an essential role in diseases, for instance, hypertension, Alzheimers disease, oxidative stress, sperm maturation and fertility, intraocular pressure, bone metabolism, fibrillary glomerulonephritis, among others. This book describes the role of ACE as a part of the Renin-Angiotensin-System, on different metabolic processes related to some diseases, such as the examples already mentioned. Moreover, ACE is related to adipose tissue. The modulation of ACE can modulate hypertension and diseases related to this enzyme. Captopril is the first successful ACE inhibitor in the treatment of hypertension; however, it has adverse effects such as dry cough, dizziness, lightheadedness or loss of taste and others. The search for alternatives to Captopril has increased research on bioactive peptides. Then, several book chapters describe how ACE can be modulated by bioactive peptides, which are short amino acid sequences previously encrypted in whole proteins and liberated through the digestion process. Animal and vegetal proteins can be used as a source of bioactive peptides. Milk, milk-derived food products and lionfish are examples of animal proteins, while several legumes such as beans and peanuts are vegetable sources of bioactive peptides.
