Описание: The main function of the Leydig cells is to synthesize testosterone, but growing evidence supports the idea that Leydig cells are important in regulating the testicular immune environment. As such, Leydig Cells: Structure, Functions and Clinical Aspects discusses Leydig cells serving as immunological sentinels in the testis. Next, the authors explore the commercially available immortalized Leydig cell lines (TM3, R2C, MA-10 and MLTC-1) derived from rat or mouse and the results of studies using these cells. Additionally, the effects of the most commonly used environmental contaminants on Leydig cell function are discussed in the context of the impaired of steroidogenic pathway. The results of several studies are presented which demonstrate that lycopene supplementation improves and maintaines the normal level of testosterone by improving the expression of StAR protein and steroidogenic enzymes in the Leydig cells of polychlorinated biphenyl exposed rats.

Описание: In Calmodulin: Structure, Mechanisms and Functions, the authors consider small and poorly-studied groups of plant calcium-dependent protein kinases that directly interact with calmodulin molecules. In plants, Ca2+ activates calmodulin-like domain kinases that do not require calmodulin or phospholipids. Thus these kinases differ from both CaMK and PKC families prevalent in mammalian cells.Next, various strategies of purification of recombinant proteins using CaM-based purification systems are reviewed and discussed. Protein purification is a crucial process in biotechnology industries and life science research laboratories. Amongst these purification strategies, affinity purification has garnered a lot of attention due to its higher speed and selectivity, leading to enhanced purity in fewer steps. In the closing chapter, the authors describe atomic-level structures of Ca2+-bound CaM (Ca2+/CaM) bound to the PSD-95 N-terminal domain. The N-lobe of CaM forms a cap that binds to the N-terminus of PSD-95 and sterically blocks the palmitoylation of PSD-95 at Cys3 and Cys5. The CaM C-lobe forms hydrophobic contacts with PSD-95 residue Y12, and the Y12E mutation abolishes Ca2+-induced postsynaptic release of PSD-95.

Описание: I?-galactosidase is an enzyme responsible for catalyzing the hydrolysis of the lactose I?-1,4 linkage into I±-D-glucose and I?-D-galactose. I?-galactosidase is found in plants, animals and microorganisms. In Beta-Galactosidase: Properties, Structure and Functions, the authors discuss the main microorganisms that produce I?-galactosidase, the characteristics of the culture media, bioprocessing parameters, the most relevant downstream steps used in the recovery of microbial I?-galactosidase, as well as the main immobilisation techniques. Next, this compilation examines I?-galactosidases classification, mechanisms, characterization and applications. Recent advances in galacto-oligosaccharides production and purification are also covered, and different challenges and future perspectives are discussed. In the closing study, strains with I?-Galactosidase activity were isolated from 25 Yak Yoghourt samples collected from the Gannan pasturing area of Gansu Province. An efficient I?-galactosidase producing strain SYA2 screened out from 21 strains was identified as Enterobacter sp. by means of morphological feature observation, physiological and biochemical characteristics measurement, and 16S rDNA sequence analysis.

Описание: Motility is an inherent property of living organisms, both unicellular and multicellular. One of the principal mechanisms of cell motility is the use of peculiar biological engines flagella and cilia. These types of movers already appear in prokaryotic cells. However, despite the similar function, bacteria flagellum and eukaryote flagella have fundamentally different structures. Chapter One of this book by Drs. Meijiao Wang, Li Zhang and Hanna Li is devoted to a comparative analysis of the flagella of prokaryotes and eukaryotes. Its believed that flagellum in eukaryotic cells appeared for the first time in ancient flagellates. In Chapter Two, Drs. P. Huitorel, M. Cachon and J. Cosson summarize the results of their long-term studies of flagellum dinoflagellates. The "evolutionary invention" of the ancient flagellates proved to be so successful that the axonemal structure in flagellum has not changed for hundreds of millions of years. In the third chapter, Drs. V. Bondarenko, G. Prokopchuk and J. Cosson reported the analyses of kinetic characteristics of flagella motions in fish using the most modern methodologies. In Chapter Four, Drs. R. Uzbekov, A. Garanina, J. Burlaud-Gaillard and C. Bressac described spermiogenesis of the parasitic wasps Cotesia congregata. Their data showed that significant shortening of the flagella occurs during the formation of the mature spermatozoon and this flagella has been the shortest spermatozoon flagella described in the animal kingdom until now. Centriole in this spermatozoon is transformed into a special structure known as the "cogwheel structure" that does not contain microtubules. In the process of multicellular organism evolution, flagella, which ensure the mobility of individual cells, gave origin to two types of cilia. Motile cilia in the tissues like cerebral ventricles, respiratory epithelium and oviducts moved liquid flows relatively to immobile cell layers. The second type of cilia (primary cilia) lost motility function and acquired cellular sensitivity function. Moving and sensitive cilia grow from basal bodies, which originate from centrioles. Chapter Five by Drs. I. Alieva, C. Staub, S. Uzbekova and R. Uzbekov discusses which of the centrioles mother or daughter creates the moving cilium and sensitive cilium. In Chapter Six, Drs. D. Conkar and E. Nur Firat-Karalar describe in detail the biochemical aspects of primary cilium assembly pathways, intraflagellar transport and ectosome release. This chapter provides an overview of the trafficking pathways involved in ciliary compartmentalization and describes the primary ciliary as a sensitive "cells antenna", participating in many regulatory processes in the cell.Defects in flagella and cilia cause many hereditary diseases. Different examples of ciliopathy are described in Chapter Seven by Drs. E. Bragina, E. Blanchard and R. Uzbekov.A wide review of experimental models, organisms and analytical methods to study flagella and cilia are presented to readers who want to learn about their main research directives.
